冠菌素对不同钾水平下TTC法测定的棉花根系活力的影响

摘 要:

冠菌素可以提高棉花根系还原氯化三苯四唑(TTC)生成三苯甲臜(TF)的能力, 本文研究了不同钾水平下冠菌素处理后的TF值在不同提取方法之间的差异及其与根系多酚和呼吸速率之间的关系。结果显示, TF热提取法的测定值明显高于常温提取法, 尤其是冠菌素处理条件下。冠菌素处理后TF的OD485值明显高于OD520值, 和冠菌素显著提高根系中花青素含量具有一致性。冠菌素处理显著提高了煮沸根和烘干根的TTC还原量, 与显著提高根系多酚含量具有一致性。冠菌素显著提高了新鲜根系的TTC还原量, 如其单位质量鲜根的OD520值分别是高钾和低钾的5.4和5.9倍(热提取法)或6.1和4.1倍(常温提取法), 同时显著提高了根系多酚含量及单位质量的呼吸速率, 这表明TTC还原能力可以用于表示冠菌素对根系代谢活性的促进, 而不受多酚的影响。

Effect of Coronatine on Cotton Root Activity Determined by TTC Assay at Different Levels of Potassium

Abstract:

Coronatine can improve the 2,3,5-triphenyltetrazolium chloride (TTC) reduction to triphenylformazan (TF) in cotton root. In this paper, we studied the differences of TF value in the different extraction methods with COR treatment under the different levels of potassium and their relationship between root polyphenol and respiration rate. The results showed that TF values of hot extraction were obviously higher than those of room temperature extraction, especially with COR treatment. The absorbance values at 485 nm with COR treatment were obviously higher than those at 520 nm, which was the same with anthocyanin contents increased by COR. And COR treatment significantly increased the TTC reduction of boiled and dry root, in accordance with polyphenols contents enhanced by COR. COR treatment significantly enhanced TTC reduction of fresh root. Compared with high K and low K respectively, OD520•g-1 (FW) were 5.4 and 5.9 times (hot extraction method) or 6.1 and 4.1 times (room temperature extraction), and significantly improved the polyphenol content and root respiration rate. Therefore, TTC reduction can be used to indicate metabolic activity increased by COR independent of polyphenols contents.
